花椒离体愈伤组织诱导研究

摘    要:以“凤县大红袍”花椒嫩叶为外植体,研究不同种类和浓度的植物生长调节剂、不同盐浓度的培养基以及同一低温不同处理时间对花椒愈伤组织诱导及其生长的影响。结果表明,MS为最适培养基;诱导花椒愈伤组织较优的生长调节剂组合为,0.5 mg/L 6-BA+0.5 mg/L 2,4-D,诱导出的愈伤生长旺盛,质地松散,无褐化现象;在0℃处理2、4 h仍可以获得正常的愈伤组织,不影响再分化。

Abstract:The tender leaves of “Fengxian Dahongpao”,a cutivar of Zanthoxyhum bungeanum were used as explants to study the effect of different factors on callus induction and the growth of Zanthoxyhum bungeanum.The results showed that MS was the best medium.The preferable combination of growth regulators was 0.5 mg/L 6-BA+0.5 mg/L 2,4-D,in which the calli grew vigorously with loose texture,and without browning.Furthermore,normal callus could still be obtained when treated 2 and 4 h in 0℃,indicating low temperature treatment did not affect re-differentiation.
