林麝生长激素基因的克隆与生物信息学分析

摘    要:了解林麝生长激素(GH)基因序列结构特点,为进一步研究GH基因的结构功能遗传变异规律及其与生长性能的关系提供科学依据。根据GenBank上已公布的绵羊全基因组GH基因序列设计合成4对特异性引物,以林麝基因组DNA为模板进行PCR扩增,将所获得的序列拼接,获得林麝GH基因CDS区序列,采用ExPASy等分子生物学工具对林麝GH蛋白的理化性质、二级结构、蛋白功能等进行预测。结果表明,林麝GH基因CDS区全长657 bp(GenBank登录序列号为KU296865),编码218个氨基酸。α-螺旋结构和卷曲结构构成了林麝GH蛋白二级结构的骨架,为跨膜蛋白,同时第1~26位氨基酸为信号肽,第27~218位氨基酸残基为生长激素结构域。进化分析结果表明,林麝与羊的关系最为接近,在分子水平上符合物种进化理论。本研究成功克隆了林麝GH基因CDS区序列,其序列保守性强,为下一步林麝GH基因的表达调控、进化和多态性分析奠定了基础。

Abstract:To reveal the structure characteristics of Moschus berezovskii growth hormone (GH) gene and provide scientific basis of the genetic variation and the relationship with growth performance of GH gene.According to the genome sequence of GH gene of the published Ovis aries from NCBI,we designed four-pair specific primers.And PCR amplification was conducted using M.berezovskii genome DNA as template.After splicing,we obtained the CDS sequence of M.berezovskii GH gene.Then,the structure and features,including physical-chemical properties,secondary structure,and function of GH protein were analyzed by ExPASY molecular biology tools.The full CDS sequence of forest musk deer GH gene was 657 bp (GenBank Accession No.KU296865),encoding 218 amino acids.Alpha helix and coil structure constituted the framework of forest musk deer GH protein secondary structure.It was a transmembrane protein.And the signal peptide was located in the 1-26 amino acids,and the growth hormone domain was located in the 27-218 amino acid residues.According to the theory of evolution at the molecular level,phylogenetic analysis showed that the relationship between forest musk deer and sheep was most close.This study successfully cloned CDS sequence of forest musk deer GH gene,with a strong conservation,which could provide the foundation of expression regulation,evolution and polymorphism analysis of forest musk deer GH gene.
