黄萎病侵染后棉花根部内生真菌群落结构分析

摘    要:为研究黄萎病菌侵染对棉花根部内生真菌群落结构的影响,采用PCR-DGGE技术分析黄萎病侵染后苗期、蕾期、花期、铃期健康棉株和发病棉株内生真菌群落组成。结果显示,在棉花生长中后期,发病棉株样品中各指数均高于健康棉株,说明黄萎病菌侵染可提高内生真菌群落结构的多样性;在DGGE电泳图谱中,对差异明显的17个条带进行切胶回收、克隆测序,发现有7个条带分布在半知菌亚门,占41.18%,4个条带分布在子囊菌亚门,占23.53%,其他条带分别为fungal endophyt、uncultured fungus、Dothideomycete和Acremonium cellulolyticus。说明:黄萎病菌侵染能明显改变内生真菌群落结构多样性。

Abstract:In order to understand the diversity of the structure of endophytic fungi communities in cotton infected with Verticillium.The aim of the study is to analyse the structure of endophytic fungi communities in health and disease cotton roots infected with Verticillium at seeding tage, bud stage, flowering stage, and boll setting stage by the technology of PCR DGGE.The results showed that the indexes of disease cotton samples were higher than the health ones at the middle late stages of cotton.The diversity of endophytic fungi communities were enhanced after cotton infected with Verticillium.Sequencing of 17 DGGE bands demonstrated that some group belonged to deuteromycotina ( 41.18% ), some group belonged to ascomycotina (23.53%), the others were fungal endophyt, uncultured fungus, Dothideomycete and Acremonium cellulolyticus, respectively.The results showed that the diversity of the structure of endophytic fungi communities in cotton was influenced apparently by cotton Verticillium wilt.
