摘    要:为保存和扩繁牡丹杂交系种质资源,提供牡丹组培苗的菌根化供试体系,本试验筛选牡丹杂交系‘Z1-10-3’和‘Z1-WM’的种子,取种胚进行胚培养,建立了牡丹胚培养无菌体系。结果表明:成熟种子经70%酒精2 min与0.1%HgCl 20 min表面消毒后去外种皮切开胚乳直接取胚接种,3 d时启动率达76.7%,比直接用种子和去外种皮的种子接种启动时间提前30~50 d。胚根苗直接培养的最佳培养基和培养条件为:1/2MS+IBA 1.0 mg/L +AC 0.4 g/L+Sugar 30 g/L,在自然弱光照下培养。

Embryo-culture of 2 Hybrid Strains Seeds of Tree Peony

Abstract:In order to conserve and multiply germplasm resources of hybrid strain of tree peony, and provide tissue culture seedling with arbuscular mycorrhizae, the germ-free system of embryo-culture was constructed in hybrid seeds of tree peony('Z1-10-3','Z1-WM').Firstly, the seed surface of tree peony were sterilized by 70% alcohol 2 min and 0.1% HgCl2 20 min. Secondly, the seed coat and endosperm were wiped off. Then the embryo was used to cultivate. About 76.7% of embryos were germinated after 3 d. The starting time of growth of explants with embryo taken directly was 30-50 d prior to the seeds with exopleura. Results showed that the best media and cultivation condition of radicle plantlet was 1/2MS+ IBA 1.0 mg/L+AC 0.4 g/L+Sugar30 g/L with natural low light.
