大叶金花草多糖的提取、分离纯化及结构分析

摘    要:采用热水提取法从大叶金花草中提取大叶金花草多糖,用酶法和Seveg法去除蛋白质,H2 O2脱色,经DEAE-纤维素(Cl -)柱层析纯化后,纸层析和Sephadex G-100 凝胶柱层析证实为单一组分,并用凝胶色谱法、薄层层析、乙酰化GC-MS、紫外光谱和红外光谱等对其组成和结构进行了研究.大叶金花草多糖平均分子质量为8.6×104 u,由阿拉伯糖、葡萄糖和糖醛酸组成,在260和 280 nm处有多糖无核酸和蛋白质的吸收峰.大叶金花草多糖是含有β-吡喃葡萄糖的非蛋白类单一多糖.

Abstract:Water-soluble polysaccharide in Stenoloma chusanum(Linn.)Ching is extracted by means of hot water.Polysaccharide is purified by enzyme treatment and Seveg method to remove protein,then decolorized by 10 % H2O2 to remove pigment,followed by DEAE-cellulose column chromatography.Polysaccharide is shown to be a single homogeneous component by PC and Sephadex G-100 column chromatography,and their structural composition was studied by means of permeation chromatography(GPC),TLC,GC-MS,UV,IR,etc..The average molecu...
