向日葵抗坏血酸过氧化物酶的电子克隆和生物信息学分析

摘    要:利用电子克隆技术得到长753 bp的向日葵抗坏血酸过氧化物酶(HaAPX)cDNA。通过生物信息学分析发现,HaAPX在核酸水平和蛋白质水平上都与常见物种的APX非常相似,具有极其保守的酶活作用域Peroxidase-1(-divtlsggHtl-)和Peroxidase-2(-aplmlRlawHsa-),且配体结合位点和金属结合位点完全保守。HaAPX与甘薯(Ipomoea batatas)APX亲缘关系很近,其编码蛋白由250个氨基酸组成,分子量为2.7 ku,无信号肽和跨膜区,二级结构由37.6%α-螺旋、13.2%β-折叠、9.2%β-转角和40%无规则卷曲组成,三维构象与豌豆APX的非常相似。

Abstract:Using in silicon cloning,the cDNA of ascorbate peroxidase(APX) was cloned from Helianthus annuus with the full length as 753 bp.The bioinformatic analysis showed that the HaAPX shared a high similarity with that of other species in DNA and protein level,possessing the domains of Peroxidase1(-divtlsggHtl-) and Peroxidase2(-aplmlRlawHsa-),as well as the ligand-binding sites and metal-binding sites were extremely conserved.HaAPX was closer to APX of Ipomoea batatas in the phylogenetic tree.HaAPX encoded a protein of 250 amino acids with the molecular weight as 2.7 ku.The signal peptide and trans-membrane region were not found in the protein sequence.The protein contains 37.6% of α-helix,13.2% of β-sheet,9.2% of β-turns and 40% of random coil.The 3-D conformation of HaAPX protein was in accordance with APX of Pisum sativum.
