摘    要:[目的]选育高纤维素酶活的绿色木霉菌株。[方法]以一株绿色木霉F1为出发菌株,以每一轮诱变处理筛选到的酶活最高的突变株作为下一轮诱变的出发菌株。经过紫外线、亚硝基胍(NTG)、硫酸二乙酯(DES)诱变处理。计录菌落数,计算致死率并测定CMCase的酶活。[结果]紫外线处理200 s时,孢子的致死率接近100%。U1突变株的相对酶活最高,为出发菌株F1的110.4%。NTG处理50 min时,孢子的致死率接近100%,N4诱变菌株的相对酶活最高,为出发菌株F1的128.9%。DES处理50 min时的诱变致死率接近100%,D8菌株的配对酶活最高,相对酶活为出发菌株F1的140.6%。[结论]得到了一株产酶量高且性状稳定的高产绿色木霉菌株D8,其CMCase相对酶活较出发菌株F1提高了40.6%。
