转基因抗2,4-D棉花室内快速鉴定方法初探

摘    要:通过在培养基中添加卡那霉素和2,4-D,研究其对抗2,4-D转基因(tfdA)棉花品系508和常规棉花品系中9409种子萌发和生长发育的影响,初步建立了一种不受时间限制的简便快速的室内鉴别转基因材料的方法。结果表明,卡那霉素最明显的特点是使非转基因材料子叶失绿,而转基因材料则呈正常绿色;2,4-D显著抑制非转基因材料的种子萌发和幼苗发育,但对转tfdA材料的影响则不大,其用量在2mg/L的情况下就能将转基因材料和非转基因材料区分开来。用该方法鉴别转tfdA基因杂交一代材料,其结果与大田鉴定相一致。

Abstract:The effects of 2,4D and Kanamycin on the seed germination and seedling growth of transgenic line 508 and nontransgenic line 9409 were studied. The results indicated that Kanamycin could cause the cotyledon of nontransgenic line 9409 turning ablio, but transgenic line 508 growed normally. Seed germination and seedling growth of nontransgenic line 9409 were restrained remarkable by 2,4D, but for transgenic line 508 lightly, They can be distinguished remarkably when medium contain 2mg/L 2,4D. On the basis, we have developed a rapid method for identifying transgenic materials, and it was tested by the field method.
