环境激素壬基酚对鳞翅目昆虫培养细胞的影响

摘    要:壬基酚(NP)是一种最新确认的环境激素。为了探讨环境激素对鳞翅目昆虫培养细胞的影响及其利用途径,研究了NP对家蚕卵巢细胞(BmN)和苜蓿丫纹夜蛾卵巢细胞(Sf9)的影响。在TC-100培养液中添加NP,体外培养细胞,MTT法调查细胞增殖,抽提细胞DNA进行损伤调查。结果表明,0.032 mmol/L以下浓度的NP对BmN细胞增殖有促进作用(p<0.01),NP浓度继续增加则出现毒害,细胞生长和增殖受到抑制(p<0.01),死亡细胞明显增加,具有浓度效应和时间效应。NP对BmN培养细胞的LC50(72 h)为0.051 mmol/L。浓度超过0.128 mmol/L时,能诱导BmN细胞DNA链发生断裂,导致DNA损伤。NP对Sf9细胞增殖有较强的抑制作用(p<0.01),有明显的浓度效应和时间效应,培养72 h时LC50为0.047 mmol/L;在试验浓度范围内,细胞生长和增殖虽然受到抑制,但死亡细胞并无明显增加。NP对鳞翅目昆虫培养细胞的生长发育具有显著影响,有诱发DNA损伤的可能。

Abstract:Nonylphenol(NP) is one of the newly-defined environmental hormones.In order to investigate the effect of environment hormone on cell of lepidopteron and to investigate its utilization pathway,the effects of NP on ovary cells of Bombyx mori(BmN) and Autographa califobiea (Sf9) were studied.The DNA-damage of cells after purification was observed by culturing the BmN and Sf9 cells with NP-added TC-100 medium in vitro and investigating the cell multiplication with method of 3-4,5-dimethy1(thiazol-1-2-yl)]-2,5-diphenyltetrazolium bromide(MTT).The results show that NP can promote the growth of BmN cells as the concentration under 0.032 mmol/L(p<0.01).With the concentration increasing,the growth and multiplication of cells are bated(p<0.01),the number of death cells increased obviously,thus there is apparent concentration-effect and time-effect.The LC_(50)of NP(culture 72 h) on BmN cells is 0.051 mmol/L.NP can incur the break of DNA strands of BmN cells as a result of the damage when the concentration is over 0.128 mmol/L.The restrained function of NP to the multiplication of Sf9 cells is obvious(p<0.01)and there is an apparent concentration-effect and time-effect.The LC_(50)(culture 72 h) on Sf9 cells is 0.047 mmol/L.Within the locatable ambit of concentration,the growth and multiplication of cells are bated,but the number of death cells increased unobvious.NP has obvious effect on the growth of lepidopteron cells and there is the possibility of incurring the damage of DNA.
