絮凝剂高产菌株的离子注入诱变选育

摘    要:[目的]提高生物絮凝剂的絮凝活性。[方法]以絮凝剂产生菌FJ-15为出发菌株,采用低能N+注入的方法对其进行诱变处理,从突变体中筛选高絮凝活性、性能稳定的菌株,并检测其絮凝活性。[结果]N+注入剂量为2×10^14ions/cm^2时,菌株存活率较低,随N+注入剂量增加,菌株存活率呈上升趋势,当N+注入剂量为8×10^14ions/cm^2时,菌株存活率最高(75.34%),当N+注入剂量为1×10^16ions/cm^2时,正突变率较大;通过初筛试验,得到了193株正突变菌株;通过复筛试验,得到了3株高絮凝活性的菌株FJM-10、FJM-19、FJM-29,其絮凝率分别为93.81%、86.64%、83.65%,平均较原菌株的絮凝活性(64.55%)高20%左右。其中,FJM-10菌株的稳定性最好,絮凝活性最高。[结论]FJM-10菌株的最佳发酵碳源和氮源分别为葡萄糖(或麦芽糖)和蛋白胨(或尿素)。

Abstract:[Objective] The aim was to improve the flocculating activity of microbial flocculant.[Method] With flocculant-producing strain FJ-15 as the original strain,the mutagenic treatment on it was conducted by implanting low energy N+,then the strain with high flocculating activity and stable performance was screened out from the mutants and its flocculating activity was detected.[Result] When the implantation dose of N+ was 2×1014 ions/cm2,the survival rate of strains was lower.The survival rate of strains showed increasing trend with the increase of implantation dose of N+ and when the implantation dose of N+ was 8×1014 ions/cm2,the survival rate of strains was the highest(75.34%).When implantation dose of N+ was 1×1016 ions/cm2,the positive mutation rate was bigger.After primary screening test,193 positive mutation strains were obtained.And though secondary screening test,3 strains such as FJM-10,FJM-19,FJM-29 with high flocculating activity were obtained,and their flocculating rate was 93.81%,86.64%,83.65% resp.,and their average flocculating activity was about 20% higher than that of original strain(64.55%).Among which,the stability and flocculating activity of FJM-10 were the highest.[Conclusion] The optimal carbon and nitrogen resource of FJM-10 were glucose(or maltose) and peptone(or urea) resp.
