磷酸钙作为O型口蹄疫多肽疫苗佐剂的研究

摘    要:为了探索磷酸钙(calcium phosphate,CaP)作为多肽疫苗佐剂的可行性,本研究以O型口蹄疫多肽疫苗为抗原,采用共沉淀法制备CaP-多肽复合物,测定包封率。将制备好的疫苗,分别在0、14、28 d腹腔注射免疫Wistar大鼠,尾静脉采血检测口蹄疫抗体水平。结果显示,CaP可包裹一定量的O型口蹄疫多肽,包封率达到50%,免疫大鼠后油佐剂组抗体效价在14 d达到26,42 d增至210并可维持35~42 d;CaP组在21 d后陆续产生抗体,抗体水平较油佐剂低,为27~28。结果表明,CaP作为佐剂相对游离多肽可以提高O型口蹄疫多肽疫苗免疫效果,但抗体水平相对油佐剂低。

Abstract:In order to explore the feasibility of calcium phosphate (CAP) as a vaccine adjuvant, the complex of CAP and type O Foot-and- mouth disease (FMD) peptide vaccine was prepare by co-precipitation. The rate of encapsulation efficiency was detected. Wistar rats were immunized intraperitoneally with the complex at day 0, 14 and 28. Tail vein blood samples were collected to detect FMD antibody levels. As a result, a certain amount of type O FMD peptide antigen was wrapped in CaP and the rate of encapsulation efficiency reached to 50%. The average antibody titer of the rats immunized with oil adjuvanted FMD antigen was 26 at day 14 and reached to 2^10 at day 42. However, the average antibody titer of the rats immunized with CaP adjuvanted FMD antigen was detectable only at day 21 post immunization and continued to rise to 27-28. The results show that CaP as an adjuvant improved immune response of type O foot-and- mouth disease peptide vaccine but its enhancement was lower than that ofoil adjuvant.
