摘 要: | [目的]建立以槲皮素为标准品用测定北柴胡中总黄酮含量的新方法。[方法]北柴胡黄酮用超声波提取,用荧光法测定总黄酮含量。[结果]当槲皮素浓度在6.7×10-6~6.0×10-4g/L时,荧光强度和槲皮素浓度呈良好的线性关系,其线性回归方程为y=531.840 48x-3.544 48,R2=0.999 03,该方法的检出限为2.51×10-7g/L;北柴胡中总黄酮的平均含量为5.045 2μg/g,RSD=5.61%,平均回收率98.44%。[结论]该方法操作简便,准确度高,可用于北柴胡药材的质量控制。

Abstract: | [Objective]Quercetin was used as standard material to establish a new method for determining total flavanoid in Bupleurum chinense DC..[Method]The total flavones was extracted from Bupleurum chinense DC.by ultrasonic wave and deternined by spectrofluorimetry.[Result]When the quercetin concentration ranged from 6.7×10-6 to 6.0×10-4 g/L,fluorescence and quercetin concentration showed the good linear relationship,linearity relationship y=531.840 48x-3.544 48,correlativity R2=0.999 03,the detectioning limit was 2.51×10-7 g/L,The average amount of total flavanoid in Bupleurum chinense DC.was 5.045 2 ug/g,RSD=5.61%.[Conclusion]It was simple and accurate method to determine total flavanoid in Bupleurum chinense DC.with spectrofluorophotometer.The new method could be used for quality control of Bupleurum chinense DC.. |
