马铃薯不同基因型幼芽茎外植体的组织培养

摘    要:利用青薯168、青薯5号和青薯8号三个品种幼芽带节茎段和节间组织为材料,研究了马铃薯的再生体系,结果表明,愈伤组织诱导率为72.92%~92.59%,愈伤组织分化率为30.68%~51.13%,每个外植体形成的小枝梢数为2.3~9.1个,绿色小植梢生根长成健壮的小植株,故认为MS+NAA0.25mg/L+BAP1.5mg/L+GA37mg/L+蔗糖30g/L+琼脂7g/L为愈伤组织诱导培养基.MS+BAP2.25mg/L+GA37mg/L+KT2.0mg/L+NAA2.25mg/L+蔗糖30g/L+琼脂7g/L为分化培养基组成的再生体系是有效的,特别对青薯8号更为明显。

Tissue culture of bud stem explants from three Qinghai genotypies of potato

Abstract:The present article deals with in vitro induction callus and shoot regeneration of bud stem explants from three Qinghai cultivars: Qingshu168, Qingshu5 and Qingshu8. The results shown that frequency of callus induction were 72.92-92.59%, frequency of differentiation were 30.68-51.13%, number of shoot per explant were 2.3-9.1. The suitable induction medium was MS +NAAO.25mg/L+ BAP1.5mg/L+GA37mg/L+ Sucrose 30g/L+ agar 7g/L. The suitable differentiation medium was MS+ BAP2.25mg/L+ GA37mg/L+KT2.0mg/L+NAA2.25mg/L+ Sucrose 30g/L+agar7g/U
