| 摘 要： | 为了分离舍饲羊群羊痘病毒并对其进行诊断研究,对山西舍饲羊群疑似羊痘发病羊的口唇痘状痴皮组织提取物进行细胞培养以扩增病毒,对病变细胞用苏木精-伊红染色法(hematoxylin-eosin staining,HE)观察,然后进行动物回归实验和酶联免疫吸附测定(enzyme-linked immunosorbent assay,ELISA).结果表明:扩增的病毒经连续培养3代后出现规律性细胞病变;用常规HE染色可见细胞中出现由于病毒感染产生的包涵体;动物回归实验表明已分离的病毒可导致动物发生痘性病变:用ELISA 检测出现显色反应,表明样品中存在与酶标抗体发生反应的病毒抗原,样品病毒浓度为6.835 IU·L-1.实验成功诊断并分离出山西舍饲羊群羊痘,为后续研究提供了理论基础. |
