IMS-PCR对食品中单增李斯特菌快速检测

摘    要:为了建立一种快速检测食品中单增李斯特菌的方法,试验设计了Listeriolysin O、23SrRNA和hly、iap四对引物分别做双重PCR引物,并与免疫磁珠法分离LMO相结合对LMO进行快速鉴定。结果显示,这四对引物分别扩增条带为700bp,240bp,308bp,505bp,可分辨不同种属的李斯特菌和其它不同菌株。建立的IMS-PCR方法最低检测限可达到1CFU/25g(mL)食品,通过检测食品样本188份,结果与常规检测方法所得的结果一致率为100%。所以,建立的IMS-PCR方法具有极好的特异性、敏感性和稳定性,在24h内可得到准确的检测结果,具有广阔的发展和市场应用前景。

Abstract:to develop a rapid detection method to test L.monocytogenes,the experiment designed four pairs of primers to do duplex PCR and separate LMO combined with Immunomagnetic Separatio methods.The four pairs of primers are Listeriolysin O,23SrRNA,hly and iap separately.The result showed that amplified band from this four pairs of primers is 700 bp,240 bp,308 bp,505 bp.We can separate different species of Listeria and other different germ strains.The detection line can reach 1CFU/25g (mL) food using the IMS-PCR methods.Using this method to test 188 food samples,we can get the identical results compared with conventional testing methods.So the conclusion was that established IMS-PCR method has excellent specificity,sensitivity and stability.We can get correct result in 24 hours.It has broad development and market prospects.
