石栗树腋芽外植体消毒方法的筛选

摘    要:[目的]筛选石栗树腋芽外植体的消毒方法,解决石栗组织培养中大田外植体污染问题,提高石栗组织培养的成活率.[方法]设计3因子3水平的正交试验,3因子分别是自来水冲洗、0.1% HgCl2和10% NaClO3溶液;3水平是自来水冲洗时间分别为60、120、180 min,0.1% HgCl2溶液浸泡时间分别为0、8、16 min,10% NaClO3溶液浸泡时间分别为0、12、24 min.观察9种处理后石栗枝条表面消毒效果.[结果]自来水冲洗120 min,0.1% HgCl2溶液浸泡16 min,10%NaClO3溶液浸泡24 min,可使污染率降到最低,为16.67%,而诱导愈伤组织成功率达到80.00%.[结论]石栗外植体灭菌的最佳组合能在外植体的承受范围之内使污染率降低到16.67%;在低温干燥和树龄小的植株上取材,污染率较低.

Abstract:[Objective]Appropriate sterilization methods for explants of Aleurites moluccana axillary bud was selected in order to solve the pollution problem in tissue culture of Aleurites moluccna and improve survival rate of tissue-cultured Aleurites moluccana. [Method]Three factors (tap water flushing, 0.1% HgCl2 soaking, and 10% NaClO3 soaking) and three levels of (60,120, and 180 min for tap water flushing; 0, 8,16 min for 0.1% HgCl2 soaking; 0,12, 24 min for 10% NaClO3 soaking) orthogonal test was designed to observe the sterilization effect on appearance of Aleurites moluccana branch processed by nine treatments. [Result]The results showed that the best combination for explants sterilization was as follows: flushed by tap water for 120 min, soaked in 0.1%HgCl2 for 16 min, and soaked in 10%NaClO3 for 24 min. [Conclusion]The above-mentioned best combination could bring down the contamination rate to 16.67% with no serious harm to explants. Explants obtained from treelet in low-temperature and dry season had little pollution to the environment.
