| 摘 要： | 建立了远志流浸膏中远志酸含量测定的高效液相色谱法。样品经盐酸溶液水解,沉淀经水洗后用甲醇溶解,进高效液相色谱仪进行检测。色谱条件：XBridge C18色谱柱（5μm,4.6 mm×150 mm）,流动相为乙腈-0.2%磷酸溶液（40∶60）,检测波长210 nm,柱温30℃,进样量20μL。远志酸的浓度在5-500μg/mL范围内与峰面积呈良好的线性关系,线性回归方程为y=9 834x-24 784,相关系数r=0.999 9。该方法简便,灵敏度高,为远志流浸膏质量标准的研究提供了科学依据。

| Abstract: | A method of HPLC for determination of polygalacic acid in extractum polygalae liquidum was developed.Hydrochloric acid solution was used to hydrolyze the extractum polygalae liquidum sample.The hydrolysate dissolved with methanol was determined by HPLC.The liquid chromatography separation was carried on XBridge C18 column（5 μm,4.6 mm×150 mm）with acetonitrile-0.2%H3PO4（40∶60） as mobile phase and the detection wavelength at 210 nm.The calibration curve was linear in the range of 5-500 μg/mL,the equation was y = 9 834x - 24 784, r = 0. 999 9. This method was simple and sensitive, which provided the scientific evidence for the research of the quality standard of extraetum polygalae liquidum. |
