20个紫花苜蓿品种的ISSR遗传多样性分析

摘    要:采用ISSR分子标记技术,对来自美国、加拿大、澳大利亚和荷兰等4个国家的20个紫花苜蓿品种进行遗传多样性和亲缘关系分析。结果显示:从100条ISSR引物中筛选出10条带型清晰、多态性较好的引物,共扩增出75条带,其中62条呈多态性条带,多态性比率(PPB)为82.67%。20个紫花苜蓿品种的遗传相似性系数为0.60~0.87,平均为0.75,其中Algonquin与Phabulous之间的遗传相似性最高,达0.87,Sanditi,WL232和4020品种间,以及32IQ和Sequel品种间的遗传相似性最低,均为0.60。UPGMA聚类分析结果可将20个紫花苜蓿品种划分为6大类群,充分说明其具有丰富的遗传多样性。

Abstract:In this paper,the genetic diversity of 20 alfalfa (Medicagosativa)varieties from America,Canada, Australia and Netherland were studied using inter-simple sequence repeat (ISSR)molecular markers in order to provide the scientific basis for analyzing the genetic diversity and genetic relationship of alfalfa.Sixty two poly-morphic bands were amplified with 10 effective primers selecting from 100 ISSR primers,and the average per-centage of polymorphic bands was about 82.67%.The value of genetic similarity (GS)indexes among those 20 alfalfa varieties varied from 0.60 to 0.87,and the average GS value was about 0.75.The GS value between Al-gonquin and Phabulous was the highest about 0.87.But the GS value between the varieties of Sanditi and WL232,4020,between the varieties of 32IQ and Sequel was the lowest,only about 0.60,respectively.These 20 alfalfa varieties were classified into 6 groups using the UPGMA cluster analysis method.Therefore,the results showed that 20 alfalfa varieties had the higher level of genetic diversity,and which would provide a theoretical basis for the genetic characteristics of alfalfa germplasm resources and the breeding of new varieties.
