草菇染色体核型的鉴定

摘    要:利用等高锁状同源电场凝胶电泳(CHEF),获得草菇的电泳核型.综合两组脉冲电泳的电泳条件、溴化乙锭染色以及DNA杂交分析结果,草菇染色体DNA至少可分为9个谱带.这一结果和以前用光学显微镜观察草菇的染色体数目的结果相一致.以混浊酿酒酵母和粟酒裂殖酵母等的相对迁移率为基准,草菇染色体长度估计在1.32~5.4Mb之间,染色体组总长约25Mb.

Abstract:An electrophoretic karyotype of Volvariella volvacea was obtained using Contour-Clamped Homogeneous Electric Field (CHEF) gel electrophoresis. Based on the conditions of 2 sets of pulsed field gel electrophoresis, the results of ethidium bromide staining and DNA hybridization analysis with two moderately repetitive probes, it was concluded that at least 9 chromosomal DNAs were found in V. volvacea. This result was consistent with previous studies using light microscopy. The chromosome size was estimated as 1.32 Mb to 5.4 Mb based on gel migration relative to the chromosomes of Saccharomyces cerevisiae, Schizosaccharomyces pombe and Hansenula wingei strains. The total genome size of V. volvacea was estimated to be about 25 Mb.
