摘    要:[目的]探索利用鱼鳔蛋白制备抗氧化肽的最佳条件。[方法]采用胰蛋白酶、中性蛋白酶、风味蛋白酶酶解海南鱼鳔,以酶解液清除DPPH的效果为评价指标,采用单因素试验和响应面试验,研究酶解的pH、温度、时间、底物浓度、酶浓度等因素对酶解效果的影响。[结果]鱼鳔的最佳酶解条件为底物浓度6.0%,酶浓度1.6%,酶解pH 6.0,酶解时间4.1 h,酶解温度63℃;该条件下制备的鱼鳔抗氧化肽对DPPH自由基的清除率达76.06%。[结论]该研究为鱼鳔蛋白的开发利用提供了科学依据。

Study on Antioxidant Peptide Prepared by Enzymolysis of Swim Bladder Protein

Abstract:[Objective] The aim was to explore the optimum conditions for preparing antioxidant peptide by using swim bladder protein.[Method] The enzymolysis was carried out on Hainan swim bladder by using trypsin,neutral protease and flavored proteinase.With scavenging DPPH effect as an evaluating index,the single-factor experiment and response surface experiment were adopted to study the effects of pH,temperature,time,substrate concentration,enzyme concentration on enzymolysis.[Result] The optimum enzymolysis conditions were substrate concentration 6.0%,enzyme concentration 1.6%,pH 6.0,time 4.1 h,temperature 63 ℃.Under the above conditions,the scavenging rate of swim bladder antioxidant peptide to DPPH was up to 76.06%.[Conclusion] The study will provide scientific basis for the development and utilization of swim bladder protein.
