大丁草愈伤组织及试管苗培养的研究

摘    要:[目的]研究大丁草愈伤组织及试管苗的培养条件。[方法]以大丁草花柄为外植体,采用组织培养方法,进行愈伤组织诱导和分化培养、不定芽的继代分化增殖培养以及试管苗生根培养、移栽和定植的研究。[结果]花柄愈伤组织诱导培养的理想培养基为MS+0.5 mg/L 6-BA+2.5 mg/L 2,4-D;愈伤组织分化培养和不定芽继代分化增殖培养的的理想培养基为MS+0.8 mg/L AgNO3+0.1 mg/LNAA+1.2 mg/L6-B;将继代分化增殖培养不定芽用浓度为1.2 mg/L IAA溶液处理24 h,再接种到1/2MS+0.1 mg/L NAA的培养基上进行生根培养的方法,是不定芽生根培养的理想方法;试管苗移栽成活率为92.8%,定植成活率为97.7%。[结论]定植成活的试管苗保持了野生大丁草的所有植物学特征,且生长旺盛,可用于进行培养繁殖。

Study on Callus and Tube Seedlings Culture of Leibnitzia anandria

Abstract:[Objective] To study the culture conditions of Leibnitzia anandria callus and tube seedlings.[Method] With the flower stalks of Leibnitzia anandria as explants,the method of tissue culture was adopted to investigate callus induction and differentiation,differentiation and multiplication of adventitious buds,rooting and transplantation of tube seedlings.[Result] MS+0.5 mg/L 6-BA+2.5 mg/L 2,4-D was the best medium for induction of callus.The optimum medium for the differentiation of callus as well as the differentiation and multiplication of adventitious buds was MS+ AgNO3 0.8 mg/L+NAA 0.1 mg/L+6-BA 1.2 mg/L;the best rooting culture method of tube seedlings was inoculating the adventitious buds into 1/2MS+ NAA 0.1 mg/L after being treated with 1.2 mg/L IAA for 24 h;the transplanting survival rate was 92.8% and stable planting survival rate was 97.7%.[Conclusion] The tube seedlings survived retained all biological characteristics of the wild Leibnitzia anandria with exuberant growth,and could be used for culture.
