摘    要:以蒙古扁桃幼龄实生苗单芽茎段为试材,以MS为基本培养基,研究了外植体消毒方法、生长调节剂对初代培养、增殖培养和生根培养效果的影响。结果表明,70%乙醇10s加0.1%HgCl21min消毒效果良好,外植体污染率18.75%,成活率81.25%;MS 6-BA 1.0 mg.L-1 NAA0.2 mg.L-1培养基初代培养,外植体萌芽率为72.2%;MS 6-BA 2.0 mg.L-1 NAA0.2 mg.L-1培养基增殖培养效果最佳,增殖率2.37;1/2MS IBA1.0 mg.L-1培养基上发根最好,14 d时生根率为40%,平均发根量3.5,平均根长2.09 cm。

Abstract:Young seedling shoots of Prunus mongolica were used as explants,using MS medium as basic medium.The explant sanitization and the effect of growth reg ulators in the induction,multiplication and rooting were studied.The results showed that shoots were treated by 0.1% HgCl2 for 1 min.after 70% ethanol for 10 seconds,effect was the best,rate of contamination was 18.75%,rate of survival was 81.25%.The suitable medium for the bud induction from shoot was MS+6-BA 1.0 mg·L-1+NAA 0.2 mg·L-1,the germination rate of bud could reach 72.2%.The optimum medium of multiplication was MS+6-BA 2.0 mg·L-1+NAA 0.2 mg·L-1,rate of multiplication was 2.37.The optimum medium of rooting was 1/2MS+IBA 1.0 mg·L-1,rooting rate was 40% after 14 days,mean number of roots was 3.5,mean length of roots was 2.09 cm.
