摘    要:以纳豆杆菌为出发菌株,进行紫外诱变,采用玉米蛋白粉为底物的平板分离法初筛及摇瓶发酵玉米蛋白粉复筛的筛选策略,以复筛发酵物中可溶性蛋白含量和抗氧化活力为指标进行筛选。结果表明,菌悬液在照射时间180 s,菌浓度为1×105个/mL下,突变株的浓醪玉米蛋白粉发酵液的可溶性蛋白质量浓度为(12.74±0.12)mg/mL,抗氧化活力达到(725.70±13.23)U/mL,突变株遗传性能稳定。

Abstract:Bacillus natto is conducted by ultraviolet mutagenesis.Mutant strain is successfully screened by plate isolation with corn gluten meal(CGM) and fermentation in flask,whose criteria are contents of soluble protein and antioxidative activity.The result indicates that under mutagenesis time of 180 s and strain concentration of 1×105 numbers/mL,the contents of soluble protein and antioxidative activity of mutant strain reached(12.74 ±0.12) mg/mL and(725.70 ±13.23) U/mL,respectively.And mutant strain shows excellent genetic stability.
