罗布麻叶总黄酮提取工艺研究

摘    要:目的]对罗布麻叶总黄酮提取工艺进行了研究。方法]采用分光光度法测定罗布麻叶总黄酮的含量,采用单因素考察不同提取方法和不同提取条件对罗布麻叶总黄酮提取率的影响,利用正交试验进一步验证,得出罗布麻叶总黄酮最佳提取工艺。结果]罗布麻叶总黄酮在波长510 nm处有最大吸收峰,线性回归方程为:C=0.2019×A-0.0100,相关系数r=0.9997;各种提取因素对罗布麻叶总黄酮提取率的影响各不相同,其中影响程度由大到小依次为:提取溶剂>料液比>提取时间>提取温度。结论]最佳提取工艺条件为:料液比1∶10,乙醇浓度65%,提取温度70℃,提取时间2 h。

Abstract:Objective] The aim was to study on extraction technology of total flavonoids from Apocynum venetum Linn.Method] The content of total flavonoids from A.venetum was determined by ultraviolet spectrophotometry.In the lab experiment with single factors the effect of different extraction methods and conditions on the extraction rate of total flavonoids from A.venetum was investigated,and then through the further verification in orthogonal experiment the optimum extraction technology of total flavonoids from A.venetum was obtained.Result] The total flavonoids in A.venetum exhibited the maximum absorption peak at wavelength of 510 nm,with the linear regression equation of C=0.201 9×A-0.010 0 and the correlation coefficient of 0.999 7.The effect of each extraction factor on the extraction rate of total flavonoids in A.venetum was different,with its effecting degree in order as the extraction solvent>solid-liquid ratio>extraction time>extraction temperature.Conclusion] The optimum extraction technology was 65% alcohol,solid-liquid ratio of 1∶10,extracting at 70 ℃ for 2 h.
