过氧化物酶UTLIEF电泳对苏丹草、高粱及其杂交种品种的真实性鉴定研究

摘    要:本试验采用超薄层等电聚焦电泳(UTLIEF),检测了苏丹草、高粱以及高粱苏丹草杂交种3个种共15个品种的幼苗、幼根以及萌发种子和干种子的过氧化物酶同工酶。结果表明:采用pH范围为4—5的载体两性电解质制备凝胶进行电泳得到的过氧化物酶电泳图谱谱带数目多且清晰度高,易于鉴定;萌动种子和干种子的过氧化物酶电泳后没有谱带出现。幼苗幼根均出现一定数目的谱带,其中以14d幼苗的过氧化物酶电泳图谱最适宜用作参试品种的鉴定。

Abstract:The POD of young seedling, young root, budding seed and dry seed of 15 varieties from sorghum bicolor, sorghum sudanens and their hybrids was examined by UTLIEF. The results showed that Ampholyte which pH range was 4-5 was suitable for identification; after electrophoresis budding seeds and dry seeds had not any bands, but the POD of young seedling and young root had some bands at the same time, the POD of young seedling for 14 days of the tested varieties was most effective for identification.
