毛叶海棠(Begonia cathayana Hemsl)的组织培养和体细胞胚的发生

摘    要:取毛叶海棠的叶片作外植体,接种于8种IAA、2,4-D、6-BA浓度配比不同的MS培养基上,45d后,含有IAA10mg/L和6-BA1ml/L的培养基上获得了胚状体。此培养基即为毛叶海棠胚状体诱导的最佳培养基。解剖观察表明胚状体源于叶表皮单细胞的垂周分裂。毛叶海棠胚状体的诱导需要适当浓度的IAA和6-BA相互配合。相同的外植体在相同诱导条件下既可发生愈伤组织又可发生胚状体,主要是由于不同细胞的生理状态不同和位置效应所致。

Abstract:The leave cuttings of Begonia cathayana Hemsl was inoculated in 8 kinds of MS medium with different proportion components of IAA,2,4-D and 6-BA. After 45 days, the embryoids of Begonia cathayara Hemsl were initiated in the medium with 10mg/L IAA and 1mg/L 6-BA,and this medium wsa selected as the best medium for the embryoid formation. The embryo initiated from some single cells of the epidermis. Appropriate proportion of IAA and 6-BA is essential for inducing the embryo of Begonia cathayana Hemsl. The reasons that both the callus and the embryo were initiated from the same cutting by the same conditions are the different physiological state and the location information of the single cells.
