摘    要:目的]为了研究柄海鞘体内的微生物能否产生具有抗菌功能的活性物质。方法]从烟台海域的柄海鞘中筛选到一株细菌,命名为N2。采用透明圈法进行抑菌试验,同时进行菌落形态、常规生理生化研究和细菌鉴定系统测试。为了进一步确定菌株N2的分类学地位,测定其16S rRNA基因序列,与相关细菌种属相应序列的同源性进行比较,并构建系统发育树。结果]菌株N2具有抑制霉菌生长的特性;菌株N2属于革兰氏阳性菌,杆状,具有鞭毛,菌落呈乳白色,菌株N2与Bacillus属细菌的特征相似;菌株N2与枯草芽孢杆菌的亲缘关系最近,相似性达99%。结论]菌株N2可鉴定为枯草芽孢杆菌,可以将其作为潜在的海洋有益菌应用于抗肿瘤和植物病害的防治。

Identification of a Marine Bacteria Producing Bioactive Substances

Abstract:Objective]The research aimed to study whether the microorganism in Styela clava could produce the active substance which possessed the antimicrobial properties.Method] A bacterium N2 was screened from Styela clava in Yantai maritime space.The bacteriostasis function of N2 in the partial mold was explored by circle clear zone.The phylogenetic analysis results basically consisted with morphologic observation and analysis results.Based on the phenotypic properties of morphology,physiology and biochemical,the identification of bacteria was made almost thoroughly and systemically.Taxonomical status of this bacterium was further demonstrated by sequencing 16S rRNA gene of strain N2,comparing with those of related strains and constructing a phylogenetic tree using neighbour-joining method.Result] N2 had strong antibacterial activity to the growth of mold.The bacterium was gram-negative which was rod-shaped and had flagellum.The microflora was opaque ivory,spore growed at secund strain N2 was similar to Bacillus in the most of the phenotypes.Strain N2 was most relative to Bacillus subtilis,and the homology of 16S rRNA gene between them was 99%.Conclusion] Strain N2 was identified as Bacillus subtilis.It can be used in intensive aquaculture as biologicals using to anti-tumor and prevent disease of plants.
