苗药铁筷子挥发油对大鼠佐剂性关节炎的作用及机制研究

目的]观察苗药铁筷子挥发油对佐剂性关节炎大鼠的作用,并初步探讨其作用机制。方法]设立正常对照组、模型对照组、阳性药物组和铁筷子挥发油高、中、低剂量组(生药量10.0、5.0和2.5 g/kg);除正常对照组外,其余各组于大鼠左后跖皮下注射弗氏完全佐剂建立佐剂性关节炎模型;各组于造模当天开始给药,连续给药21 d,观察铁筷子挥发油对佐剂性关节炎大鼠模型关节肿胀、免疫器官脏器系数以及病理形态的影响,采用放免法测定铁筷子挥发油对各组大鼠血清中白介素-1β(IL-1β)和肿瘤坏死因子-α(TNF-α)含量的影响。结果]与模型组比较,铁筷子挥发油对大鼠足肿胀有明显的抑制作用,对免疫器官没有明显影响;铁筷子挥发油还可明显降低佐剂性关节炎大鼠血清中IL-1β以及TNF-α的含量。结论]铁筷子挥发油对佐剂性关节炎大鼠具有一定治疗作用,其机制可能与减少细胞因子的生成有关。

Effects of Essential Oil from Chimonanthus praecox in Guizhou on Rat Adjuvant Arthritis and Their Mechanisms

Objective] To study the effects of essential oil from Chimonanthus praecox on rat adjuvant arthritis and their mechanism.Method] Rats were divided randomly into normal control group,model control group,positive drug group and high-,middle-and low-dosage groups of essential oil from Chimonanthus praecox in Guizhou(10.0,5.0 and 2.5 g/kg crude drug).Adjuvant arthritis(AA) rat model were established by subcutaneous injection of Freunds adjuvant(FCA) except the normal group.Drugs were given by oral gavage for 21 d continuously from the modeling day.The anti-arthritic effects and mechanism of essential oil from Chimonanthus praecox in Guizhou against RA were studied through observing the paw swelling,viscera index pathological changes and levels of interleukin 1β(IL-1β) and tumor necrosis factor α(TNF-α) after medication.Result] The essential oil from Chimonanthus praecox in Guizhou could effectively inhibit the articular swelling,compared with model group.But it had no significant effects on the immune organs.The contents of IL-1β and TNF-α in the serum of AA rats were decreased significantly compared with model group.Conclusion] The essential oil from Chimonanthus praecox in Guizhou had certain therapeutic effects on rat adjuvant arthritis,and its mechanism might be related to the reduction of cytokine generation.
