玉米胚性愈伤组织诱导与分化的影响因素

摘    要:以玉米自交系黄早4、综31、金黄96B、GY237、478及海921为材料,通过改良培养基上附加成分的组成及浓度,对影响玉米胚性愈伤组织的诱导及分化等几个关键因素进行了研究。结果表明,基因型对玉米胚性愈伤诱导的影响是显著的,而且基因型和基本培养基之间存在互作;在培养基上附加水解酪蛋白(casein hydrolysate,CH)、L-脯氨酸(L-proline)对于玉米胚性愈伤组织的诱导分化是必要的,而附加L-谷氨酰氨、甘露醇、AgNO3对提高胚性愈伤组织的诱导作用不显著;结果还发现胚性愈伤组织诱导培养基和胚性愈伤组织分化培养基的组合对成苗率有明显的影响作用。

Abstract:In this article, young embryo of maize inbred lines huangzao 4,zong 31,jinhuang 96B,GY237, 478, and hai 921 were cultured in vitro and some important factors affecting maize embryogenic callus induction and differentiation were studied. The results indicated that genotype has remarkable effect on embryogenic callus induction, moreover there is interactive relation between genotype and base medium. Supplementary CH(casein hydrolysate)and L-proline was requisite to embryogenic callus induction on medium. L-Glutamine,inositol,mannitol,AgNO3 had little effect on embryogenic callus induction. The combination of embryogenic callus induction and differentiation base medium had a large effect on embryogenic callus differentiation.
