秋水仙碱诱变绿玉树多倍体研究

摘    要:采用培养基添加法、浸泡法以及先预培养后处理法对绿玉树丛生芽进行多倍体诱变.结果表明:在培养基中添加0.2%的秋水仙碱进行14d培养以及先预培养3d再用0.2%秋水仙碱进行24h处理法可以获得较好的诱变效果.其成活率和诱变率分别达到53.13%、60.00%和82.35%、72.22%.茎、叶、气孔、保卫细胞、染色体以及过氧化物酶等形态生理指标的检测证明四倍体较二倍体发生了显著变化.

Abstract:Caespitose buds of Euphorbia tirucalli L.were treated with colchicine in different ways to induce polyploidy. Satisfactory results were achieved when the buds were cultured on a medium supplemented with 0.2% colchicine for 14 days or they were pre-culture for 3 days and then transferred to a 0.2% colchicines-containing medium and culture for 24 h. The survival rate of the explants and their induction rate was 53.13% and 60.00%, and 82.35% and 72.22%, respectively in the two treatments. Compared with the diploids, remarkable changed were found to have occurred in the morphological indicators of stem, leaf and guard cell and the physiological indexes of chromosome and peroxidase in their tetraploid counterparts.Caespitose buds of Euphorbia tirucalli L.were treated with colchicine in different ways to induce polyploidy.
