涡旋法提取白僵菌孢子DNA技术研究

摘    要:[目的]试验探索一种利用涡旋法提取白僵菌(Beauveria)孢子DNA的简易方法;[方法]对常规的DNA提取方法进行改进,从白僵菌孢子中提取DNA,对白僵菌孢子量、石英砂、裂解液、涡旋时间、NaAc 5个因素进行正交优化试验;[结果]获得的DNA提取体系为白僵菌孢子量0.02 g、石英砂1.0 g、裂解液2 mL、涡旋时间5 min、NaAc 200μL,其余同常规;[结论]该方法具有需用孢子量少、时间短、DNA得率高等优点。

Abstract:This study developed a simple and efficient method to extract DNA from spores of Beauveria by a vortex method.The improved methods which by five factors such as spores,quartz sand,lysate,vortex time,NaAc were used to extract the DNA of Beauveria spores,through orthogonal optimization test,an optimal DNA extraction system was obtained:Beauveria bassiana spores 0.02 g,quartz sand 1.0 g,lysate 2 mL;the mixtures were eddied for 5 min at first,then 200 μL NaAc was added,the rest procedures were the same with the conventional method.The method has the advantages of fewer spores required,time shortened,and more DNA obtained.
