小麦未熟胚离体培养的研究——愈伤组织发生及器官建成

摘    要:对小麦胚离体培养过程中愈伤组织发生及器官的形态建成进行了解剖学观察。结果表明,胚龄对愈伤组织发生的起始部位有明显影响。胚的各个部位对离体培养反应的敏感程度随胚龄的增长盾片形成愈伤组织的能力而减弱,3周龄至成熟胚的盾片不能产生愈伤组织,只起分泌α-淀粉酶、转化、吸收和运转养分的作用。同时还观察到输导系统分化完善之前存在传递细胞。不定根及不定芽的分化和形成,既存在器官发生途径,也存在胚状体途径。根、芽原基形成的先后顺序并不固定。

Abstract:The study deals with the anatomy of callus induction and organogenesis of wheat em-bryo in vitro culture.The study results indicated that theinitial part of callus induction was af-fected by the age of embryos used.Different parts of embryo varied in reacting to culture invitro.The ability of scutellum to initiate callus decreased along with the increase of embryoage.The scutella of 3-week-aged or older embryo couldn't produce any callus but secrete α-amylase,absorb,transform,and transfer nutrient from the edosperm.Transfer cells were ob-served before the establishment of conducting system.The differentiation and fromation ofadventitious root and shoot could be from either organogenesis or embryoids.Formationsequencies of root and shoot primordia were not consistent.
