东莨菪内酯对家蚕中肠组织GSTs活性的影响

摘    要:目的]探讨东莨菪内酯对家蚕中肠组织谷胱甘肽S-转移酶(GSTs)活性的影响,为桑园使用东莨菪内酯防治虫害的安全性及家蚕抗性品系选育提供理论依据.方法]采用毒叶法对家蚕进行添毒,测定东莨菪内酯对家蚕中肠组织GSTs活性的影响,并通过离体抑制试验求出东莨菪内酯对GSTs的抑制中浓度(IC50).结果]摄食不同浓度东莨菪内酯(0.026~0.104 mol/L)处理桑叶的家蚕表现出相似的中毒症状(吐液和拒食),但在添毒36h后中毒症状完全消失.相对于正常的家蚕,摄食东莨菪内酯处理桑叶后家蚕中肠组织的GSTs比活力呈前期降低、中期升高、后期降低的变化趋势.不同浓度(0.001~0.016 mol/L)的东莨菪内酯均显著抑制家蚕中肠组织离体GSTs活性,且抑制率随东莨菪内酯浓度的增加而增加.东莨菪内酯对家蚕中肠组织GSTs的IC50为0.00474 mol/L.结论]东莨菪内酯可抑制家蚕中肠组织GSTs活性.

Abstract:Objective]The influence of scopoletin on glutathione S-transferases (GSTs) in mesenteron of Bombyx mod Linnaeus was studied to provide references for mulberry field pest control and resistant line breeding of Bombyx mori Lin- naeus. Method]This research adopted the leaf poisoning method to add poison to B. moti Linnaeus with scopoletin to find out the influence of scopoletin on GSTs in mesenteron of B. mori Linnaeus. IC50 of scopoletin on GSTs was detected via in vit- ro inhibition test. Result]B. mori Linnaeus showed similar toxic symptoms (spitting and anti-feeding) after eating mulberry leaves that were treated with different concentrations of seopoletin (0.026-0.104 mol/L). However, symptoms completely disappeared after 36 h. Compared with the normal silkworm, GSTs activity trends of midgut of silkworm fed with mulberry leaves treated by scopoletin went down in earlier stage, up in the middle stage and down again in late stage. Scopoletin under different concentrations could strikingly suppress the GSTs specific activity in mesenteron of B. mori Linnaeus. With the in- crease of concentration of seopoletin, the suppression ratio increased. IC5o of scopoletin on GSTs was 0.00474 mol/L. Conclusion ]Scopoletin can repress GSTs activity in mesenteron of B. mori Linnaeus.
