草菇菌丝体超氧化物歧化酶

摘    要:采用 NBT 光化还原抑制技术检测8个草菇菌株的6个不同周(?)期的菌丝体超氧化物歧化酶(SOD)活性水平发现,草菇菌丝体在第2周龄期 SOD 活性最强高的可达光化还原抑制47%。低的为28%。从第3周龄期至第6周龄期 SOD 活性反复波动,(?)均在第2周龄期活力水平之下。菌丝 SOD 活性水平,可反映出草菇菌株子实体产量和生长速度水平。从不同龄期菌丝产量变化表明菌丝生长快、衰老快和自溶快等特性。采用聚丙烯酰胺凝胶电泳结果表明,参试的8个菌株6个周龄期的草菇菌丝在凝胶板上 Rf 值0.3附近有一条 SOD 同工酶强带,这条带可能属于 Mn-SOD 类型。从菌丝体 SOD 同工酶谱情况,可判断草菇子实体(?)伞的快慢。

Abstract:The NBT photochemical reduced inhibition technique was used to study the levels of superoxidc dismutase(SOD)activities in various weekly age of mycelia of the straw mush- room including eight strains.The result showed that the level of SOD activity of mycelia was stranger on second weekly age than that of the other weekly age and decreased or undulated during third up to sixth weekly age in all trials.The high yield strains were to hold either strong activity of SOD of the mycelia to start with as 25% of NBT photochemical re- duced inhibition level,and less enlarge undulate activity of SOD or strong activity of those mycelia to start with as 25% of NBT photochemical reduced inhibition level,and enlarge un- dulate activety of SOD,but retained 15% of NBT photochemical reduced inhibition levels to end yet.Whatever,early havest strains hold high strong activity of SOD of the mycelia, their NBT photochemical reduced inhibition levels were about 40%.At the process of cul- ture,the yield of the mycelia not stabilized in quantity.However,the mycelia of straw mush- room was mutured fast,senescenced quickly,and self-dissolved.The gel electropheregram of SOD isozyme indicated that one major SOD isozymic band was observed in location of all gel plates with the Rf.value near 0.3 for trial materials.There was only major SOD isozymic band at various weekly age in three strains.Additionally,the other SOD isozymic bands were distributed differnetly on slow,medial,and far regions of gel plates for another five strains.The major SOD isozymic band for the mycelia of straw mushroom could belong to the ran-SOD.The fruitbody of straw mushroom of strains V_(11) and V_(85-7) held thin outer volva(egg stage of fruitbody)was casily to break,and two SOD isozymic band were on slow region of gel plate.While,strains V_(801-1) and V_(85-12) of straw mushroom held outer volva bro- ken not easily and only one band of SOD isozyme existed on the parttern.The peak value of SOD isozymic bands was even constant profile by the scanning.
