摘    要:核受体辅激活蛋白基因(NCOA1)是类固醇激素受体超家族中的一员,在生殖细胞的生长和分化过程中起着重要的调节作用。本研究利用PCR-SSCP技术对NCOA1基因外显子的多态性进行了分析,在第3和12外显子上分别发现了SNPs位点,并采用实时荧光定量PCR对多态性位点处mRNA表达水平进行了研究,发现NCOA1基因随着生长发育其表达量呈波动性增加,14周龄前表达量很少,随着性成熟NCOA1基因表达量快速增加,28周龄时其表达量达到峰值,之后表达量下降。产蛋性能较好的母系鸡NCOA1基因的相对表达量比产蛋性能较差的父系鸡较早地开始增加,并且在14周龄后表达量显著高于父系鸡(P<0.05)。同位点对产蛋性能有显著影响的优势基因型AA和CC个体的表达量在16周后也显著高于同位点的其他基因型个体(P<0.05)。研究提示NCOA1基因mRNA表达水平影响鸡的性成熟过程和产蛋性能。

Abstract:Nuclear receptor co-activator 1 gene (NCOA1) is one of the members of steroid hormone receptor which is essential for growth and differentiation of ovarian follicles. Single nucleotide polymorphisms (SNPs) were detected by PCR-SSCP, and two SNPs was found at exon 3 and 12. Rea1-time quantitative PCR was used for identification of expression changes of NCOA1 gene at exon 3 and 12.
