摘    要:本研究运用杂交瘤技术成功地分离了B_8、I_2两株分泌抗伊氏锥虫单克隆抗体(McAb)的细胞系。首先用可溶性伊氏锥虫抗原接种BALB/C小鼠,经过一定的免疫程序后,取其脾细胞与SP2/O骨髓瘤细胞融合,经过两次有限稀释法克隆,用ELISA方法选育出B_8、I_2两株产生抗伊氏锥虫McAb的细胞株。用琼脂扩散法与标准羊抗鼠亚类血清反应证明两种McAbs皆隶属于IgG_1亚类。细胞株在连续传代及冻存复苏后其分泌抗体能力稳定;经多种血清学反应证实,这两株McAb与伊氏锥虫可溶性抗原不起凝集反应,只有ELISA反应阳性。pH稳定试验表明,B_8株McAb比I_2株稳定。

Abstract:Applying hybridoma techniques, B_8 and I_2 cell strains which secret monoclonal antibodies (McAbs) against Trypanosorna cvansi were successfully developed. Hybridoma ceils were derived by fusing spleen cells from immunized BALB/c mice with SP2/O myloma cells. For properties of these McAbs, it is identified that they are specific to T. evansi and belong to IgG_1 subcless by immunodiffusion, having no agglutination or precipitation reaction with soluble antigens of T. evansi confirmed by immunodiffnsion,counter immunoelectrophoresis,and indirect haemagglutination test. Stability experiments of McAbs to pH variation showed that B_8 McAb is more stable than I_2 whose viability degenerates evidently at pH 9 and pH 5.5. In addition, the secreting ability of two cell strains are unvariable in culture as well as after thawing out from liquid N_2 storages for one month.
