金荞麦内转录间隔区(ITS)的扩增及序列分析

摘    要:以2份金荞麦为材料,对其内转录间隔区ITS区序列进行PCR扩增,均获得长约700 bp的特异性产物。测序结果表明,金荞麦1号样品包含ITS序列660 bp,金荞麦2号样品包含ITS序列646 bp,在Genebank中的登录号分别为DQ780602和DQ780600。对现有金荞麦ITS的序列比对表明,其同源率为80.2%~93.1%,变异位点主要在ITS1,表现出较丰富的遗传多样性。采用邻接法构建了金荞麦的系统进化树。

Abstract:Species of the genus Fagopyrum dibotrys are important cereals that contain abundant nutrition and potential medical value.The ITS regions of two wild F.dibotrys accessions were amplified by PCR and sequenced directly.The ITS regions of the two species are 660 and 646 bp in length. The two sequences have been submitted to Genebank with accession numbers DQ780602 and DQ780600.The sequence similarity among eight F.dibotrys species varied from 80.2% to 93.1% and this variation occured mainly in ITS1.This suggests that abundant genetic diversity exists in F.dibotrys.A phylogenetic tree was constructed by the neighbour-joining method.Accession DQ780602 is a separate clade that differs from all other accessions.
