转基因油菜的遗传学分析

摘    要:本文从以下几方面对油菜转Bt基因植株进行了研究:R_1、R_2代的Km抗性;R_0、R_1代的Southern杂交;R_0代抗虫试验以及R_0代的酯酶活力测定.实验结果分别是:经Km抗性试验,芥菜型油菜R_1代在12个无性系中有10个无性系符合孟德尔单基因分离的遗传规律,R_2代在7个无性系中有6个符合这一规律;分子杂交证明部分油菜植株的基因组中存在Bt基因的同源顺序,表明Bt基因已整合进植物基因组;抗虫试验的结果是芥菜型油菜叶片饲喂幼虫10d后各处理组活虫体重与对照比,有5个无性系存在显著差异,其中最高虫试死亡率是77.7%,甘蓝型油菜则有9个无性系存在显著性差异,最高死亡率是70%;酯酶活力测定的结果表明外源基因没有影响酯酶活力.

Abstract:In this paper we studied the kanamycin resistance of R1and R2generations, Southern blotting annlysis of R0and R1gencrations, insect resistance of R0generation and the measurment of the total activity of non-specific esterases. The results of Km resistance were ten clones of R1and six clones of R2had the segregation rates of 3: 1. Southern blotting analysis confirmed that the genomes of some plants contained the homo geneous sequence of the Bt gene. The mortality of the larvae up to 77.7% in test for insecticidal activity of B. juncea and 5 clones were high difference in the larvae weight after ten day's feeding. In the same way, the results of B. napus'up to 70% and 9 clones' were high difference. Activities of the non-specific esterases in transgenic plants were no differ-ence with the control, which indicated that the foreign gene did not influence the activity of non-specific esterases.
