一株陈化烟叶附生细菌的筛选鉴定及其烟叶发酵效果

摘    要:以K326(B2F))陈化烟叶为材料,采用平板分离技术分离培养出46株活体纯培养物,依次编号为CHJ1~CHJ46,纯化菌株通过蛋白酶和淀粉酶鉴别培养基筛选培养,鉴别筛出CHJ3和CHJ9两株菌具有淀粉酶水解活性,CHJ5、CHJ28和CHJ36三株菌具有蛋白酶水解活性。CHJ3、CHJ5、CHJ9、CHJ28和CHJ36五株菌菌悬液烟叶发酵试验表明,CHJ5菌株促进烟叶的发酵效果最好,发酵后的烟叶化学成分更加合理,评吸得分达到91分,与其他处理差异达显著水平。对菌株CHJ5的多相鉴定结果表明,CHJ5隶属于微杆菌属(Microbacterium)菌株,暂命名为Microbacterium sp. CHJ5。菌株CHJ5作为烟叶发酵微生物制剂开发的备用菌株,应用前景广阔。

Screening and identification of epiphytic bacteria from aged tobacco leaf and its effect on tobacco fermentation

Abstract:Using K326 (B2F) aged tobacco leaf as material, 46 strains were isolated and cultured by plate separation technology, numbered as CHJ1-CHJ46 in sequence. CHJ3 and GXZY9 strains were screened for amylase hydrolyzing activity through the culture medium of protease and amylase screening. CHJ5, CHJ28 and CHJ36 had protease hydrolysis activity. The tobacco leaf fermentation test of CHJ3, CHJ5, CHJ9, CHJ28 and CHJ36 strains showed that the CHJ5 strain had the best effect on promoting the fermentation of tobacco leaves, and the chemical composition of the fermented tobacco leaves was more reasonable, and the smoking score reached 91 points, which was significantly different from other treatments. The results of multiphase identification of CHJ5 showed that CHJ5 belonged to Microbacterium, temporarily named Microbacterium sp.CHJ5. Strain CHJ5 has a broad application prospect as a standby strain for tobacco fermentation microbial preparation.
