半边旗提取物5F对人病理性瘢痕成纤维细胞增殖的影响

目的：研究半边旗活性成分5F对人病理性瘢痕成纤维细胞增殖的影响。方法：将不同浓度5F作用于体外培养的人病理性瘢痕成纤维细胞，检测MTT及LDH值，用免疫组织化学法检测用药前后细胞核内增殖核抗原(prolife rating cell nuclear antigen，PCNA)的表达。结果：5F在10～120mg／L浓度范围对病理性瘢痕成纤维细胞呈剂量依赖性增殖抑制，LDH值在0～40mg／L浓度范围内无明显改变，PCNA的表达在5F作用后明显减弱。结论：5F在体外能明显抑制病理性瘢痕成纤维细胞增殖，浓度在40mg／L范围内不引起细胞坏死性改变。

Effect of compound 5F isolated from Pteris semipinnata L.on proliferation of pathological scar-derived fibroblasts

Objective:To study the effect of compound 5F isolated from Pteris semipinnata L. (PSL)on proliferation of pathological scar derived fibroblasts.Methods:After treated with different concentrations of 5F,inhibition rate,lactate dehydrogenase(LDH)and proliferating cell nuclear antigen(PCNA)expression of pathological scar derived fibroblasts were detected by MTT assay,automatic biochemical analysor and immunohistochemical method respectively.Results:5F had a dose dependent inhibition of fibroblast proliferation at strengths of 20 120 mg/L( P< 0.01).LDH level of fibroblasts treated with 0 40 mg/L of 5F showed no obvious change( P> 0.05),but it remarkably elevated with 80 120 mg/L of 5F( P< 0.01) .PCNA expression of fibroblasts significantly decreased after incubated with 20 80 mg/L of 5F( P< 0.01).Conclusion:Compound 5F could substantially inhibit the proliferation of pathological scar derived fibroblasts in vitro,and there is no fibroblast necrosis at 40 mg/L or less of 5F.
