以Bar为选择标记的遗传转化体系的建立

摘    要:Bar基因是转基因商品化作物中应用最多的目标基因,也是水稻遗传转化中应用较多的选择标记基因,因此,建立以Bar基因为选择标记的通用和高效的遗传转化体系非常必要。本研究中,以籼型水稻明恢63为受体,Bar基因作选择标记,采用农杆菌介导的遗传转化法,将Bar基因转入其中,获得了一批转化子,初步建立了稳定、高效的水稻遗传转化体系。随后,以此体系为基础,将装有不同基因的两个载体pBar-1C^*和pBar-1Ab进行了转化,分别从5400块和4800块愈伤组织中获得156个和115个抗性愈伤,其抗性愈伤率分别为2.8%和2.4%,分化率分别为80.7%和87.8%,由此说明,此转化体系相对稳定,且分化率高、抗性愈伤率也较高,可用于同类选择标记的遗传转化,加快水稻转基因育种步伐。

Abstract:Bar gene is widely used in commercialized transgenic crops as a selective marker and also used in rice transformation.Therefore,the establishment of a kind of common and efficiency genetic transformation system is necessary.In the study,by using indica rice Minghui 63 as transformation receptor and Agrobacterium-mediated genetic method,Bar gene was transformed into Minghui 63.Thus,a stable and efficiency transformation system of rice was founded.Subsequently,on the basis of the system,the vectors pBar-1C* and pBar-1Ab were transformed.156 and 115 of resistant calli were obtained from 5 400 and 4 800 calli,respectively.Their transformation efficiency were 2.8% and 2.4%,and differentiation rates were 80.7% and 87.8%,respectively.These results suggest the transformation system is stable as well as efficiency,and good for transformation of rice with Bar gene as selective marker.
