摘    要:为选出更高效、实用的牛胚胎性别鉴定方法,分别用成年牛血样提取的基因组DNA和冷冻的牛早期胚胎细胞DNA为样本,以电泳法和非电泳法用相应的性染色体DNA引物做胚胎PCR性别鉴定实验,以相对比。血液模板DNA结果与胚胎样本结果都表明,使用DYZ-1引物的非电泳法在比使用SE引物的电泳节约1h以上时间的同时,具有比SE更高的灵敏度。这次研究在我国首次成功地把非电泳PCR法应用到牛胚胎性别鉴定之中,减少了胚胎性别鉴定的操作步骤,并大大缩短了性别鉴定所需要的时间,从而使胚胎移植现场性别鉴定更加可行。

Abstract:In order to develop a more efficient and applicable method of embryo sexing,both genome DNA samples extracted from cattle and embryo DNA samples were used in the contrast experiments between electrophoretic and nonelectrophoretic PCR embryo sexing.Nonelectrophoretic PCR sexing using DYZ-1 primers seems more sensitive to low DNA concentration than electrophoretic PCR sexing using SE primers,meawhile it took about 1 hour less than SE.This study successfully applied nonelectrophoretic PCR to bovine embryo sexing for the very first time in China,it also reduced a traditional step in embryo sexing.It further shortened the time for sexing,therefore made PCR sexing in field more applicable.
