同羊ETAA1基因InDel检测及其与生长性状的关联分析

摘    要:为寻找同羊中与生长性状相关的分子标记,试验对ETAA1基因InDel的多态性与同羊生长性状进行了关联分析.以166只无亲缘关系的同羊为研究对象,通过Ensembl数据库筛选ETAA1基因潜在的InDel位点,利用PCR和琼脂糖凝胶电泳对筛选到的InDel位点进行检测和分型后,进一步对InDel位点不同基因型的同羊生长性状进行关联分析.结果 显示,位于绵羊3号染色体的ETAA1基因第1内含子存在一个插入12 bp的InDel突变;根据插入的类型,可以分为II,DD和ID 3种基因型;检测出的InDel位点多态性对同羊公羊臀端高有显著影响,该位点Ⅱ基因型个体臀端高显著高于ID型与DD型(P<0.05).因此,可以把ETAA1基因此位点InDel多态作为同羊臀端高性状选择的候选分子标记.

Abstract:To search for molecular markers associated with growth traits in Tong sheep, the experimental association analysis between the InDel polymorphisms of ETAA1 gene and growth traits of Tong sheep was conducted. 166 unrelated Tong sheep were selected for this study and the potential InDel loci of the ETAA1 gene were screened by Ensembl database. After detecting and typing the screened InDel loci by PCR and agarose gel electrophoresis, we performed further association analysis for growth traits of Tong sheep with different genotypes of InDel loci. The results showed that there was an InDel site with a 12 bp insertion in the first intron of the ETAA1 gene located on sheep chromosome 3; according to the type of insertion, three genotypes were classified as II, DD and ID; the detected polymorphism of the InDel had a significant effect on hip height of the raw, and the II genotype of the locus was significantly higher than that of the ID and DD genotypes (P < 0.05).Therefore, the InDel polymorphism of the ETAA1 gene can be used as a candidate molecular marker to select the hip height of the Tong sheep.
