‘云想容’海棠茎段组织培养技术

摘    要:以‘云想容’海棠当年生单芽茎段为研究对象,采用组织培养法,研究了不同消毒剂、消毒时间、不同生长素和细胞分裂素浓度对‘云想容’海棠组织培养的影响,以期建立高效快速的‘云想容’海棠组培快繁体系。结果表明:最佳的消毒方法为用75%酒精消毒30 s后,用0.1%氯化汞处理8 min,该消毒方法能明显降低外植体的污染率,对外植体造成的伤害较小,成活率高达83.3%;最佳腋芽诱导培养基为MS+6-BA 1.0 mg·L-1+NAA 0.1 mg·L-1,芽生长健壮且诱导率高达90%;MS+6-BA 0.5 mg·L-1+TDZ 0.02 mg·L-1+NAA 0.1 mg·L-1为最佳增殖培养基,增殖系数为8.63;一步生根法以培养基1/2MS+IBA 1.0 mg·L-1较好,平均根长5.26 cm,生根率为73.3%,相比较而言,两步生根法可有效促进‘云想容’海棠的生根,生根率可达91.67%。

Abstract:The effects of different disinfectant,time of disinfection,concentration of auxin and cytokinin on tissue culture of Malus‘Yunxiangrong’were studied by tissue culture method,in order to establish rapid propagation system of Malus‘Yunxiangrong’.The results showed that the best disinfection method of primary culture was 75%alcohol disinfection for 30 seconds,and treatment with 0.1%mercuric chloride 8 minutes,this disinfection method could obviously reduce the contamination rate of explants,the damage caused by external implants was small,the survival rate was as high as 83.3%.Best axillary bud induction medium MS+6-BA 1.0 mg·L-1+NAA 0.1 mg·L-1,bud growth robust and induction rate up to 90%.The one-step rooting method was better with the medium 1/2 MS+IBA 1.0 mg·L-1,with an average root length of 5.26 cm,and a rooting rate of 73.3%.In comparison,the two-step rooting method could effectively promote the rooting of the begonia,and the rooting rate could reach 91.67%.
