摘    要:为了提高杏鲍菇原生质体产量,以杏鲍菇菌株为材料,通过单因素试验研究酶浓度、酶解时间、酶解温度、菌丝培养方式、菌龄以及稳渗剂种类对杏鲍菇原生质体制备的影响。结果表明:杏鲍菇原生质体制备的最佳条件为:酶浓度2%、酶解时间2.5 h、酶解温度29℃、菌丝液体静置培养5天,以甘露醇为稳渗剂,在此条件下原生质体产量为33.4×106个/mL,为杏鲍菇原生质体技术育种提供参考。

Abstract:In order to improve the Pleurotus eryngii protoplast yield, the optimal conditions such as enzyme concentration, enzymatic hydrolysis time, reaction temperature, mycelium culture way, mycelium age and type of osmotic stabilizer for the lywallzyme hydrolysis of the cultured myeelia of Pleurotus eryngii for protoplast production were investigated by single factor experiment. The results showed that the highest production rate of the protoplast was emerged under the conditions that 5 days old liquid mycelium in the enzyme solution of 2% lywallzyme digested for 2.5 h at temperature of 29℃ with 0.6 mol/L mannitol as osmotic pressure stabilizer,and under this conditions the protoplast yield was 33.4 × 10^6 per milliliter. This study has laid the foundation of protoplast breeding of Pleurotus eryngii in the future.
