产纤维素酶低温菌株的分离鉴定

摘    要:为促进秸秆还田,加快秸秆在北方设施土壤中的降解速度,以腐烂秸秆为待筛菌株原料,以玉米秸秆粉为培养基,在15℃低温条件下,采用刚果红平板法进行初筛,得到16株菌株,对16株菌株进行酶活测定复筛,其中L-13酶活最高,达1 679.61 U/ml。对L-13菌株的菌体形态、菌落特征进行观察,并进行了一系列生理生化试验和16S rDNA序列分析,初步鉴定L-13为枯草芽孢杆菌(Bacillus subtilis)。此菌可以在15℃下快速生长繁殖并高产纤维素酶,低温条件下能快速降解秸秆。

Abstract:In order to promote straw returning and speed up its degradation in the North greenhouse soil, using rotten straw as raw materials and corn straw powder as medium,16 strains were isolated with Congo Red plate method at 15℃.Then the enzyme activities of 16 strains were detected.L-13 had the highest enzyme activity of 1 679.61 U/ml.The mycelial morphology and colony characteristics of L-13 were observed,and a series of physiological and biochemical tests and 16S rDNA sequence analysis were conducted.The L -13 strain was preliminarily identified as Bacillus subtilis.Owing to growing fastly and high yielding of cellulase at 15℃,it could degrade the straw under low temperature rapidly.
