地涌金莲不同部位外植体多酚氧化酶活性及总酚含量分析

摘    要:通过对比分析地涌金莲原变种与红苞变种外植体PPO活性及总酚含量的差异,为组织培养适宜外植体的选择提供理论依据。结果表明,地涌金莲原种与红苞变种不同部位总酚含量存在显著差异,均表现为雄蕊最高,其中原变种为21.42 mg·g~(-1),红苞变种为20.64 mg·g~(-1),且均显著高于其他部位;红苞变种叶片及叶脉总酚含量分别为9.43mg·g~(-1)和2.32 mg·g~(-1),显著高于原变种的4.67 mg·g~(-1)和0.72 mg·g~(-1),而红苞变种雌花子房壁总酚含量为4.96 mg·g~(-1),显著低于原变种的9.44 mg·g~(-1),其余8个部位的总酚含量在2变种间无显著差异;原变种叶片及叶脉的PPO活性分别为971.5和542.8(0.01△A·g~(-1)·min(-1)),显著高于红苞变种的277.2和428.5(0.01△A·g~(-1)·min(-1)),其他部位的PPO活性两者无显著差异。表明总酚含量及PPO活性均较低的花苞片、雄花子房是地涌金莲组织培养较理想外植体材料。

Abstract:Musella lasiocarpa,which belongs to a monotypic genus,is endemic to southwest of China,including Yunnan and Sichuan province.It is a precious wild flower germplasm resource known for its dignified plant type and beautiful inflorescence.The differences in phenol content and PPO activity between explants of M.lasiocarpa var.Lasiocarpa and its mutant M.lasiocarpa var.Rubribracteata were quantified in order to provide some theoretical basis for selecting appropriate explants in tissue culture.The results showed that there was a significant difference in total phenol content among different parts of the two varieties,of which the total phenol content in the stamen was the highest with 21.42 and 20.64 mg·g-1 in Lasiocarpa and Rubribracteata,respectively.The total phenol content in leaves and veins of M.lasiocarpa var.Rubribracteata was 9.43 and 2.32 mg·g-1,respectively,which was significantly higher than that in M.lasiocarpa var.Lasiocarpa (4.67 and 0.72 mg·g-1,correspondingly).Contrarily,the total phenol content in ovary walls of M.lasiocarpa var.Rubribracteata (4.69 mg·g-1) was significantly lower than that in Lasiocarpa (9.44 mg·g1).There was no significant difference among the other eight parts.For the PPO activity,the leaves and veins of M.lasiocarpa var.Lasiocarpa were 971.5 and 542.8 (0.01△A·g-1·min-1),respectively,which was significantly higher than Rubribracteata (277.2 and 428.5,correspondingly).There was no significant difference among the other parts of the two varieties in the aspect of PPO activity.It is suggested that bracts and male ovary which have relatively low total phenolic content and PPO activity are the ideal explants for tissue culture ofM.lasiocarpa.
