RAPD分析用白花丹参叶片DNA的提取方法

摘    要:本研究采用改良CTAB法和SDS法提取新鲜白花丹参叶片的基因组DNA,初步筛选RAPD扩增引物。结果表明改良CTAB法提取的DNA较SDS法质量好,随机引物p2扩增条带相对较为清晰。再利用p2随机引物对2种方法提取的DNA进行RAPD检测比较,结果显示仅改良CTAB法能扩增出有效条带,说明改良CTAB法更适合用于白花丹参基因组DNA的RAPD检测分析。本文将为白花丹参叶片DNA的提取提供方法学参考。

Abstract:In this study,we extracted the genomic DNA from the fresh leaves of Salvia miltiorrhiza Bge f.alba by using an improved CTAB method and SDS method and preliminarily screened RAPD amplification primers.The results showed that the DNA quality of improved CTAB method was better than SDS method.On the other hand,the amplification bands of random primer 2 was clearer than others.Then we carried out RAPD assays comparison between these two methods with random primer 2,the result displayed that we got the efficiency fragments only by the improved CTAB method,which illuminated that the improved CTAB method was more suitable for RAPD assays analysis for the genomic DNA of Salvia miltiorrhiza Bge f.alba.So the improved CTAB method was more proper for genomic RAPD assay of Salvia miltiorrhiza Bge f.alba.Consistent with these results,this paper would provide a methodology reference for extracting the genomic DNA from the fresh leaves of Salvia miltiorrhiza Bge f.alba.
