多羟基双萘醛(WCT)抗血清的制备与不同酶联免疫吸附检测法的对比试验

摘    要:通过对多羟基双萘醛分子式上的醛基的改造,利用混合酸酐法使小分子化合物多羟基双萘醛能够与大分子蛋白质BSA相结合,形成人工抗原,注射兔子,成功制备WCT抗血清。通过ELISA检测,血清的效价为1∶256,非专化性效价为1∶2。本试验比较了酶联夹心法和间接酶联法的灵敏度,发现间接法检测WCT的灵敏度比夹心法相对高,这为应用间接法检测WCT提供了依据。

Abstract:The manual antigen was prepared by modified the aldehyde of WCT and linking the small molecular compound-WCT and the larger molecular protein-BSA by the way of mixing acid anhydride.Then injected into the rabbit to prepare the antiserum of WCT.The titer of antiserum and nonspecific reached to 1:256 and 1:2 respectively by ELISA detection.Compared the sensitivity of enzyme-linked sandwich assay and indirect enzyme-linked assay,the latter is more applicable.This provided the evidence to detect WCT using the indirect enzyme-linked assay.
