铁皮石斛高效再生体系研究与应用

摘    要:以原球茎为实验材料,对影响铁皮石斛再生体系的主要影响因素进行试验,建立铁皮石斛在生产中的高效再生体系。试验结果表明,在基本培养基MS、Fonnesbech和N6中,最适合的基本培养基为MS;原球茎最佳增殖培养基为MS+6-BA0.75mg/L+NAA0.2mg/L+4.0%白糖,接种35d后原球茎增殖倍数为4.19;最佳分化培养基为Ms+6-BA0.3mg/L+3.0%白糖,分化率为74%。分化出的丛芽在生根培养基MS+AC0.05%+白糖3.0%中,28d生根率达到87%。

Abstract:Protocorms of Dendrobium officinale were used as materials to determine main factors affecting the regeneration system for D. officinale, based on which a highly efficient regeneration system for D. officinale was developed. The results showed that the best basic culture medium was MS among MS, Fonnesbech (1972) and N6 (1974) tried; that the best multiplication culture medium was MS+6-BA0.75 mg/L+NAA0.2 mg/L+4.0% white sugar, with multiplication rate of the protocorms being 4.19 times 35 days after inoculation; that the best differentiation culture medium was MS+6-BA0.3 mg/L+3.0% white sugar, with 74% of differentiation rate; and that differentiated cluster shoots had a rooting rate of 87% when cultured on the rooting medium (MS+AC0.05%+white sugar 3.0%) for 28 days.
